How to Measure Absorbance with a UV-Vis Spectrophotometer

  1. Warm up and select wavelength: Switch on, allow the lamps to stabilise, and set the analytical wavelength (it is set automatically).
  2. Blank the instrument: Fill a clean quartz cuvette with the reference solution and zero the absorbance (100% T).
  3. Insert the sample: Place a 10 mm cuvette with your sample in the compartment.
  4. Read the result: Record absorbance or %T on the LCD, or send the data to the PC through the USB interface.

Frequently Asked Questions

What applications is this UV-Vis spectrophotometer used for?
It is used for pharmaceutical analysis (drug assay, dissolution testing, impurity profiling), water testing, environmental monitoring, food testing, and life science work such as protein estimation and enzyme kinetics.
What is the difference between single beam and double beam UV-Vis spectrophotometers?
A single beam instrument passes light through one path, so you measure the blank and sample in turn. A double beam instrument splits the light to monitor sample and reference together, which lowers drift. Single beam models are simpler and more economical, and suit routine analysis.
What type of light source does this system use?
It uses a deuterium lamp for the ultraviolet range and a tungsten halogen lamp for the visible range, covering 190 to 1000 nm.
What is the wavelength range and bandwidth?
The wavelength range is 190–1000 nm with a 2 nm spectral bandwidth, wavelength accuracy of ±1.0 nm and photometric accuracy of ±0.5% T.
Is it compatible with standard cuvettes?
Yes. The sample compartment takes standard 10 mm path length cuvettes, including quartz cuvettes for UV measurements.
Can I measure DNA, RNA or protein with it?
Yes, in cuvette volumes, for example A260 for nucleic acids and A280 or colorimetric assays for protein. For 1–2 µL samples without a cuvette, see the EzDrop 1000 or EzDrop 1000C microvolume spectrophotometers.
Does it come with software and data storage?
It has an LCD display, internal data storage and USB connectivity with a PC interface for software-based analysis and reporting.
Which laboratories benefit most from this model?
Its economical, compact design suits college and university teaching labs, pharma QC labs, hospital diagnostic labs, water testing labs and contract testing facilities.
What is the Beer-Lambert law and why does it matter here?
The Beer-Lambert law states that absorbance is proportional to concentration and path length. A UV-Vis spectrophotometer uses it to convert measured absorbance into concentration.
What is the UV-Vis spectrophotometer price in India?
Price depends on configuration and quantity. Submit a quotation request to receive a price list along with AMC, installation and training details from Global Bioscience Solutions, Bengaluru.
Do you offer installation, training and AMC?
Yes. We provide installation, user training, warranty and AMC service support across India.
Can this be supplied for tenders and institutional purchases?
Yes. We supply to universities, government research facilities and pharma companies and provide institutional quotations on request.
